There is a site I go to that I told safari (never) to remember the settings to (ie username and password) autofill. However, now I would like to have it autofill and I can't find how you tell safari to allow it again.

Go in the Safari Prefs under AutoFill... If you click on edit it normally will list your autofill info for all sites.

To follow up on that thought, simply delete the site that you previously didn't want filled. When you go back to the site, it should then prompt you if you want to remember the info.
